چکیده

The work was aimed to develop a method for extraction and identification of the marker compounds of Hypericum species, hyperforin and hypericin present in the buds and leaf extracts of Hypericum hookerianum, a lesser known species of Asia. LC-MS hyphenated techniques were used and the data indicated the presence of both these active compounds in the methanol extract of both buds and leaves. ABTS (2, 2 ́azinobis [3-ethylbenzthiazoline]-6-sulfonic acid) radical cation scavenging assay using the extract showed concentration dependent increase in inhibition indicating free radical scavenging and antioxidant properties. Further, the extract (500μg) showed a broad spectrum antibacterial activity especially against the drug resistant Bacillus subtilis, Staphylococcus aureus, Klebsiella pneumonia and Salmonella typhimurum which is characteristic of hyperforin. Hyperforin and hypericin when docked with the protein tyrosine kinase C using bioinformatics tools predicted anticancer potential of the extract to be validated by wetlab experiments. The results suggest methanolic extract of Hypericum hookerianum is a valuable commodity with potent antibacterial and anticancer activities. Though the shoot extracts of Hookerianum are reported to be antiHSV-1, the extracts subjected to P-24 antigen assay for HIV screening in the present investigation did not yield positive results upto 300 μg.

LC-MS and LC-PDA analysis of Hypericum empetrifolium and Hypericum sinaicum.

Within the framework of our continuous efforts to explore Hypericum species from Jordan, we report the analysis of the major active metabolites, naphthodianthrones and phloroglucinols, in the methanolic extracts of two under-explored Hypericum species; H. empetrifolium Willd. and H. sinaicum Hochst. & Steud. ex Boiss., using LC-(+,-)-ESI-MS (TIC and SIM) and LC-UV/Vis spectroscopy. Based on the...

Gc-ms Analysis and Anticancer Activity of Methanol Extract of Leaves of Hypericum Hookerianum Wight & Arn

Objective: The present investigation was an attempt to study the anticancer activity of the methanol extract of the leaves of H. hookerianum. GC-MS analysis revealed that the methanol extract of leaves contained important secondary metabolites viz. oxygenated hydrocarbons and phenolic hydrocarbons. These phytochemicals w e r e p r e s u m e d responsible for various pharmacological actions like...

Improvement of Hypericin and Hyperforin Production Using Zinc and Iron Nano-oxides as Elicitors in Cell‏ Suspension Culture of St John's wort (Hypericum perforatum L.)

Zinc and iron nano-oxides (100 ppb) were promoted the hypericin and hyperforin production in Hypericum perforatum cell suspension culture. High performance liquid chromatography method was used for detectection and identification of hypericin and hyperforin in H. perfuratum cell suspension cultures elicited with different concentrations of zinc and iron nano-oxide (0, 50, 100 and 150 ppb) after...

Monoamine Oxidase-A Inhibition and Associated Antioxidant Activity in Plant Extracts with Potential Antidepressant Actions

Monoamine oxidase (MAO) catalyzes the oxidative deamination of amines and neurotransmitters and is involved in mood disorders, depression, oxidative stress, and adverse pharmacological reactions. This work studies the inhibition of human MAO-A by Hypericum perforatum, Peganum harmala, and Lepidium meyenii, which are reported to improve and affect mood and mental conditions. Subsequently, the an...

Hyperforin accumulates in the translucent glands of Hypericum perforatum.

BACKGROUND AND AIMS Hypericum perforatum contains the therapeutically important compounds hypericin and hyperforin. Hypericin is known to accumulate in the dark glands. This investigation aimed to determine the accumulation site of hyperforin. METHODS Dark and translucent glands as well as non-secretory tissue in leaves were manually isolated under the microscope. Hyperforin content was quant...
